No, only SOME hybrid mixtures are unable to reproduce – in general most closely related species are inter-fertile, only a minority are mules. Recent research (last 30 years) has shown that Neanderthals were inter-fertile with homo sap.

I’ll guess that the knife is a test. If more than one person arrive at this double-door, and each one is greedy for the prize, well, suppose it takes at least two to open those two doors (not just each holding an Aegis against it). But the knife might lead to just one surviving greedy person at the doors, and thus it stays closed.
